The cleared supernatant was treated with formalin (final concentration 0.005%) for 48 h. Next, 30 ml of the inactivated virus was loaded on 5 ml of a 20% sucrose cushion per centrifugation tube (Beckman, SW28 tubes). After 2 h centrifugation with 104,000 × g the supernatant was discarded and resulting pellets were pooled in Tris buffered saline (TBS). An aliquot of the resulting vaccine preparations was subjected to a safety assay to exclude any possible remaining infectivity.
